B cell development is tied to selection of H and L chains; B cell development occurs in bone marrow, dependent on stromal cells and soluble factors (e.g. Cytokines and growth factors); development w/in bone marrow occurs independent of Ag; after mature B cell leaves bone marrow, circulates for several days (if it doesnt encounter Ag, dies; if it encoutners Ag, activated and differentiated); to survive first stages of development, B cells must make a productive rearrangement of the H chain and L chain genes or cell dies by apoptosis. Stimulate Ab production in absence of T cell help; B cell activation doesn't result in memory; sometimes result in isotype switching but less than by TD Ag; Ab produces are less specific and less numerous; required second signal for full activation via IFN-gamma produced by innate immune cells; 2 different type of TI Ag (TI-1, TI-2): TI-1 = mitogens, result in non-specific activation of B cells and can activate both naive and memory B cells of any specificity, TI02 can only activate Ag-specific mature B cells.
